Abstract: A protocol for extraction of good quality , high molecular weight genomic DNA from young leaves of coconut was standardized using two detergents viz. sodium dodecyl sulphate (SDS) and hexadecyl trimethyl ammonium bromide (CTAB) at different concentrations; at pH 8.0 and pH 9.0. SDS (1 %) at pH 8.0 was found to yield good quantity of high quality DNA. The extracted DNA was amenable to PCR and restriction digestion.
